The word itself literally means "the desire to discern".

A common pericope found in DN 2, DN 25, and MN 39, among other places, gives a clue:
Abandoning doubt, he dwells with doubt left behind; without uncertainty as to what things are wholesome, his mind purified of doubt.

K.R. Norman translates it as "exertion", with alternate translation (from either I.B. Horner or Ven. Walpola Rahula) as "tenacity".

When the mind is concentrated (unified), phenomena become clear, i.e., one can see them in detail (e.g., their arising, passing, necessary conditions).

I agree with Mike. Perhaps the letting go also refers to letting go of involvement with what's occurring in the present -- not getting hooked by these events, but just watching them come and letting them go by.
